Antiviral drug, non-competitive non-nucleoside HIV-1 reverse transcriptase inhibitor. Nevirapine binds to reverse transcriptase and blocks the RNA-dependent and DNA-dependent DNA polymerase activities by causing a disruption of the enzyme's catalytic site. It does not inhibit HIV-2 transcriptase and human DNA polymerases α, ß, γ, or δ. It reduces the viral load and CD4 counts when used in combinations with other antiretroviral drugs. Resistant virus emerges rapidly and nearly always when nevirapine is administered as monotherapy. Cross-resistance to other non-nucleoside reverse transcriptase inhibitors is observed in vitro.
