Antiviral drug, nucleotide reverse transcriptase inhibitor. Tenofovir in converted in vivo to tenofovir, a nucleoside phosphonate (nucleotide) analog of adenosine monophosphate. Subsequently, it is converted to tenofovir diphosphate, the active metabolite, that inhibits HIV-1 reserve transcriptase by incorporating into the viral DNA chain and inducing the chain termination. Tenofovir is a weak inhibitor of mammalian DNA polymerases α, β, and mitochondrial DNA polymerase γ.
