In addition to affecting DNA replication, chlorambucil induces cell apoptosis by accumulation of the cytosolic protein p53 with subsequent activation of the apoptosis promoter (Vach). The cytostatic effect of chlorambucil is due to both chlorambucil itself and its main metabolite - phenylacetic acid mustard.
